Pregnenolone, 3-beta-hydroxy-5-pregnen-20-one, is a natural steroid hormone produced in the body from cholesterol. It has been described as the “mother of all steroid hormones” — all adrenal cortical steroid hormones, of which there are over 150, are derived from it.

Since many of these hormones tend to decline with age, deficiency can lead to increased cholesterol synthesis by the liver. Studies show the body makes around 60% less of these adrenal hormones at 75 than it does at 35. One rarely discussed contributor to cholesterol levels in older adults, besides diet, is a deficiency of pregnenolone. “Adrenal fatigue syndrome”, a pattern seen fairly commonly in older people under chronic stress, can result from its depletion with age.

Pregnenolone has been linked to positive support of the immune system, mood and memory in ageing adults. In the body it can be converted into progesterone and other mineralocorticoids, as well as DHEA and other sex steroids. The body is likely to convert pregnenolone according to its needs, replenishing other adrenal cortical hormones. One of these is the glucocorticoid cortisol, which helps maintain healthy blood glucose levels; pregnenolone is a precursor in that pathway.
